Apolipoprotein B (Apo B) is a major protein constituent of chylomicrons (Apo B-48), LDL (Apo B-100) and VLDL (Apo B-100). ApoB-100 (Apolipoprotein B100) is the primary protein in low-density lipoprotein (LDL) cholesterol. It is the largest of the ApoB group of proteins, consisting of 4563 amino acids. Importantly, there is one ApoB100 molecule per hepatic-derived lipoprotein. Apo B-100 functions as a recognition signal for the cellular binding and internalization of LDL particles by the apoB/E receptor. Mutations (changes) in apoB100 can cause a condition called familial hypercholesterolemia. ’s ApoB100 (Human) ELISA Kit is used for the quantitative detection of ApoB100 in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of ApoB100 in the sample.
