Aquaporin 4 (AQP4) is a water channel protein that belongs to the aquaporin family of the proteins. AQP4 is abundantly expressed on the perivascular astrocyte endfeet. In addition to the brain, spinal cord, and optic nerve, AQP4 is also expressed on the epithelial cells of other organs such as the kidney, skeletal muscles, intestines, and salivary glands. One of the main functions of AQP4 is the bidirectional transport of water and maintain water homeostasis in the central nervous system (CNS). Additional functions include synaptic plasticity, potassium homeostasis, memory formation, and astrocyte migration. Nearly 80% of the patients suffering from a rare inflammatory demyelinating CNS disorder called Neuromyelitis Optica (NMO) contain antibodies to AQP4 in their blood. Studies using animal models have also demonstrated that AQP4 may play a role in diseases such as cerebral edema, ALS, epilepsy, Alzheimer’s, and Parkinson’s disease. ’s AQP4 (Human) ELISA kit quantitatively measures AQP4 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of AQP4 in the sample or standard.
