Argininosuccinate synthase 1 (ASS1) is an ATP-dependent rate-limiting enzyme in the arginine biosynthetic pathway. ASS1, together with arginosuccinate lyase, catalyzes the conversion of L-citrulline and aspartate to arginosuccinate. ASS1 gene mutations are observed in Citrullinemia type I, a rare autosomal recessive disorder that causes neonatal hyperammonemia, increased citrulline levels, and early neonatal death. Reduced levels of ASS1 have been observed in multiple human cancers, such as pancreatic, liver, breast, bladder cancer, glioblastoma, and melanoma, and correlates with poor prognosis in patients. ’s ASS1 (Human) ELISA kit is based on the Sandwich principle to quantitatively measure the concentration of ASS1 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is directly proportional to the concentration of ASS1 in the sample or standard.
