Protein Standard


A protein standard, also known as a protein ladder or molecular weight marker, is a mixture of proteins of known sizes used as a reference in gel electrophoresis experiments. Protein standards are essential tools for estimating the size of unknown proteins and for determining the molecular weight of protein samples separated on gels.

Here's how protein standards are used in gel electrophoresis experiments:

Size Determination: Protein standards consist of a mixture of proteins with known molecular weights, typically ranging from a few kilodaltons (kDa) to several hundred kilodaltons. These proteins are run alongside the protein samples of interest on a gel during electrophoresis.

Molecular Weight Calibration: By comparing the migration distances (or migration rates) of the protein standards to the migration distances of the protein samples, researchers can estimate the molecular weight of the unknown proteins. This is achieved by plotting a standard curve with the known molecular weights of the protein standards against their migration distances.

Normalization: Protein standards are also used for normalizing gel-to-gel variability and assessing the efficiency of protein transfer in techniques such as Western blotting. Including a protein standard lane on each gel allows researchers to compare protein band intensities across different gels and experiments.

Quality Control: Protein standards serve as quality control measures to ensure the proper functioning of electrophoresis equipment and reagents. They allow researchers to verify the resolution and accuracy of protein separation and to identify any issues with gel integrity or experimental conditions.

Protein standards are available in various forms, including pre-stained or unstained, colored or colorless, and ready-to-use or concentrated formats. They may also include a mixture of proteins with different fluorescent or colorimetric labels to facilitate detection and visualization on gels.

Commonly used protein standards include:

  • Benchtop Molecular Weight Markers: These are mixtures of proteins with known molecular weights that can be run on standard polyacrylamide gels.
  • Pre-stained Protein Ladders: These protein standards are pre-stained with dyes for visualization on gels without additional staining steps.
  • Fluorescent Protein Ladders: These protein standards are labeled with fluorescent dyes for detection and quantification using fluorescence imaging systems.
