Description: ADAM10 is a member of the ADAM (a disintegrin and metalloproteinase) gene family and have been implicated in a variety of biological processes involving cell–cell and cell–matrix interactions, including fertilization, muscle development, and neurogenesis. It is Involved in the cleavage of the adhesion molecule L1 at the cell surface and in released membrane vesicles, suggesting a vesicle-based protease activity. It also controls the proteolytic processing of Notch and mediates lateral inhibition during neurogenesis. ADAM10 has been identified as the constitutive α-secretase in the process of amyloid-β protein precursor (AβPP) cleavage and plays a critical role in reducing the generation of the amyloid-β (Aβ) peptides. New treatments and preventive interventions targeting ADAM10 regulation for Alzheimer's disease are necessary, considering the wide variety of ADAM10 substrates. ’s ADAM10 (Human) ELISA Kit is used ntitative detection of ADAM10 in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of ADAM10 in the sample.
