Disintegrin and metalloproteinase domain-containing protein12 also known as ADAM12 belongs to the ADAM (a disintegrin and metalloprotease) protein family. ADAM12 is expressed in placenta, skeletal, cardiac, and smooth muscle. ADAM12 is involved in skeletal muscle regeneration, specifically at the onset of cell fusion and also involved in macrophage-derived giant cells (MGC) and osteoclast formation from mononuclear precursors. They are also expressed in some tumor cells lines. Additionally, it binds insulin growth factor binding protein-3 (IGFBP-3) and appears to be an effective early Down syndrome marker. ADAM12 has also been implicated in the development of pathology in various cancers, hypertension, liver fibrogenesis and asthma. ’s ADAM12 (Human) ELISA Kit is used for the quantitative detection of ADAM12 in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of ADAM12 in the sample.
