Vimentin is a type III intermediate filament protein that is expressed in macrophages and mesenchymal cells. Some of the major functions of vimentin include maintaining the shape of the cell, cytoplasmic integrity, and stabilizing cytoskeletal interactions. Vimentin also plays a role in cell migration and proliferation and is implicated in the initiation and progression of cancer. Vimentin undergoes citrullination by peptidyl arginine deiminases during the epithelial-to-mesenchymal transition, apoptosis, and tissue inflammation and hence invokes an autoimmune inflammatory response. Antibodies to mutated citrullinated vimentin (MCV) have been detected in patients with autoimmune diseases such as rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE). Hence, anti-MCV antibodies serve as efficient biomarkers for the diagnosis of RA and SLE. ’s anti-MCV (Human) ELISA kit quantitatively measures anti-mutated citrullinated vimentin antibodies in human serum, plasma, and other biological fluids. The kit is based on the principle of Sandwich ELISA. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is directly proportional to the concentration of anti-MCV in the sample or standard.
