Description: Bone morphogenetic protein 3 (BMP-3) also known as osteogenin belongs to the transforming growth factor beta superfamily. BMP-3 plays an essential role in developmental process by inducing and patterning early skeletal formation and by negatively regulating bone density. It antagonizes the ability of certain osteogenic BMPs to induce osteoprogenitor differentiation and ossification. BMP-3 initiates signaling cascades by associating with type II receptor ACVR2B to activate SMAD2-dependent and SMAD-independent signaling cascades including TAK1 and JNK pathways. It is an antagonist to other BMPs in the differentiation of osteogenic progenitors. It is highly expressed in fractured tissues. BMP-3 is hypermethylated in many cases of colorectal cancer (CRC) and hence along with other hypermethylated genes, may be used as a biomarker to detect early-stage CRC. ’s BMP-3 (Human) ELISA Kit is used for the quantitative detection of B-3 in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of BMP-3 in the sample.
