Description: Bone morphogenetic protein 6 (BMP-6) belongs to the transforming growth factor-β (TGF-β) signaling family and has a very broad biological role in development. It plays an essential role in many developmental processes, including neurogenesis, vascular development, angiogenesis and osteogenesis. BMP-6 is able to induce all osteogenic markers in mesenchymal stem cells. It plays also an important role in the regulation of iron metabolism by acting as a ligand for hemojuvelin/HJV. It initiates the canonical BMP signaling cascade by associating with type I receptor ACVR1 and type II receptor ACVR2B. In turn, ACVR1 propagates signal by phosphorylating SMAD1/5/8 that travel to the nucleus and act as activators and repressors of transcription of target. BMP-6 has a role in chondroid formation, and also tubulo-glandular differentiation in pleomorphic adenomas. ’s BMP-6 (Mouse) ELISA Kit is based on the Sandwich-ELISA principle. Ticro-ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse BMP-6. Standards or samples are added to the micro-ELISA plate wells that bind to the specific antibody. Then a biotinylated detection antibody specific for Mouse BMP-6 and Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. The wells are washed, a TMB substrate solution is added to the wells and blue color develops in proportion to the amount of BMP-6 bound. The enzyme-substrate reaction is terminated by the addition of stop solution and turns color. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm. The OD value is proportional to the concentration of Mouse BMP-6. The concentration of Mouse BMP-6 in the samples can be calculated by comparing the OD of the samples to the standard curve.
