Fibroblast growth factor 21 (FGF-21) is a pleiotropic hormone that belongs to the FGF family of proteins. Liver is the primary source of FGF-21; however, it may also be present in other tissues such as the skeletal muscle, white adipose and brown adipose tissue, and pancreas. FGF-21 expression is regulated by PPARα in the liver, whereas in the adipose tissue, the expression is regulated by PPARγ. FGF-21 is a master regulator of energy homeostasis. Under fasting conditions or ketogenic dietary intake, FGF-21 expression increases and it promotes hepatic gluconeogenesis to produce glucose to other tissues. It stimulates the uptake of glucose only in the adipose tissue but not any other cells. Overexpression of FGF-21 in transgenic mice protects mice from diet-induced obesity and lowers glucose and triglyceride levels in the blood of diabetic rodents. Increased levels of FGF-21 were reported in the serum of patients suffering from Type II diabetes and non-alcoholic fatty liver disease (NAFLD). Thus FGF-21 could serve as an efficient therapeutic target for the treatment of metabolic diseases. ’s FGF-21 (Human) ELISA kit quantitatively measures FGF-21 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of FGF-21 in the sample or standard.
