FOLR1 also known as Folate receptor alpha belongs to the folate receptor (FOLR) family which has high affinity for folate and folic acid analogs at neutral pH. Exposure to slightly acidic pH after receptor endocytosis triggers a conformation change that strongly reduces its affinity for folates and mediates their release. FOLR1 is predominantly expressed on epithelial cells and is dramatically upregulated on many carcinomas and acts as a potential biomarker and therapeutic target for medulloblastoma. It plays a vital role in folate transport across the blood–brain barrier through the choroid plexus. It is also essential for neural tube closure during embryogenesis. ’s FOLR1 (Human) ELISA Kit is used for the quantitative detection of FOLR1 in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of FOLR1 in the sample.
