Hemoglobin is involved in oxygen transport from the lung to the various peripheral tissues. 's Hemoglobin ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in the kit has been pre-coated with an antibody specific to human Hemoglobin. Standards or samples were added to the micro ELISA plate that reacts with biotinylated detection antibody specific for human Hemoglobin and Avidin-Horseradish Peroxidase (HRP) conjugate which on the addition of substrate form blue color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm. The OD value is proportional to the concentration of human Hemoglobin. The concentration of human Hemoglobin in the samples can be calculated by comparing the OD of the samples to the standard curve.
