Test Principale: This assay employs a two-site sandwich ELISA to quantitate ADAM10 in samples. An antibody specific for ADAM10 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyADAM10 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ADAM10 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ADAM10 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: A Disintegrin And Metalloprotease 10 is a ADAM family member that cleaves many proteins including TNF-alpha and E-cadherin. Members of the ADAM family are cell surface proteins with a unique structure possessing both potential adhesion and protease domains. ADAM10 is a sheddase, and has a broad specificity for peptide hydrolysis reactions. New data on the ADAM10 shedding of ephrin/eph complex associated with Eph on one cell surface has determined that ADAM10 cleaves ephrin, within the ephrin/eph complex, formed between two cell surfaces. When ephrin is freed from the opposing cell, the entire ephrin/eph complex is endocytosed. This shedding in trans had not been previously shown, but may well be involved in other shedding events.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
