Application: This Human Chordin (CHRD) ELISA Kit employs a two-site sandwich ELISA to quantitate CHRD in samples. An antibody specific for CHRD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCHRD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CHRD is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CHRD bound in the initial step. The color development is stopped and the intensity of the color is measured.

• Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
• It is recommended that all samples and standards be assayed in duplicate or triplicate.

Background: Chordin is a polypeptide that dorsalizes the developing embryo by binding ventralizing TGFβ proteins such as bone morphogenetic proteins. It may also play a role in organogenesis. There are five named isoforms of this protein that are produced by alternative splicing.In humans, the chordin peptide is encoded by the CHRD gene. 948-amino acid protein containing 4 internal cysteine-rich repeats and 4 potential N-glycosylation sites. Northern blot analysis detected high levels of a 3.7-kb Chrd transcript in 7-day postcoitum mouse embryos; its level decreased at later developmental stages and in adult tissues. Dot blot analysis of mRNA expression in human adult and fetal tissues showed that CHRD is expressed at the highest level in liver.
