Test Principale: This assay employs a two-site sandwich ELISA to quantitate XPO5 in samples. An antibody specific for XPO5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyXPO5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for XPO5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of XPO5 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: Exportin-5 belongs to a large family of karyopherins that mediate the transport of proteins and other cargo between the nuclear and cytoplasmic compartments. The deduced 1, 205-amino acid protein has a calculated molecular mass of about 136 kD. XPO5 and XPO1 share 25% amino acid identity. Northern blot analysis detected transcripts of about 3.6 and 5.0 kb in all tissues examined.XPO5 bound the nucleoporins NUP214 and NUP153 in a RAN-GTP-independent manner and bound a cargo protein, ILF3, in a RAN-GTP-dependent manner. XPO5 bound ILF3 at its double-stranded RNA-binding domain (dsRBD) and also bound the dsRBDs of other proteins. XPO5 directly interacted with VA1 RNA in a RAN-GTP-dependent manner.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
