Test Principale: This assay employs a two-site sandwich ELISA to quantitate PLXNB3 in samples. An antibody specific for PLXNB3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPLXNB3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PLXNB3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PLXNB3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: Tamagnone et al. (1999) identified the cDNA sequence of a novel member of the plexin gene family, which they called plexin B3.Artigiani et al. (2004) showed that mammalian Plxnb3 is a high-affinity semaphorin-5A (SEMA5A) receptor. Plxnb3 was activated by Sema5a, and the Sema5a-Plxnb3 complex triggered intracellular signaling through Met, the receptor for hepatocyte growth factor.Tamagnone et al. (1999) found the PLXNB3 coding sequence in the genomic sequence of the ABCD1 gene on Xq28.Matsumoto et al. (2005) identified a partial deletion (exons 3 to 10) of the ABCD1 gene with fusion to the neighboring PLXNB3 gene. Sequencing showed that the breakpoints were at nucleotide +1374 in intron 2 of the ABCD1 gene and nucleotide +915 in intron 2 of the PLXNB3 gene.
