Test Principale: This assay employs a two-site sandwich ELISA to quantitate POGK in samples. An antibody specific for POGK has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPOGK present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for POGK is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of POGK bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: The exact function of the protein encoded by POGK is not known. However, this gene product contains a KRAB domain (which is involved in protein-protein interactions) at the N-terminus, and a transposase domain at the C-terminus, suggesting that it may belong to the family of DNA-mediated transposons in human. The Krueppel-associated box (KRAB) is a domain of around 75 amino acids that is found in the N-terminal part of about one third of eukaryotic Krueppel-type C2H2 zinc finger proteins (ZFPs) . It is enriched in charged amino acids and can be divided into subregions A and B, which are predicted to fold into two amphipathic alpha-helices. The KRAB A and B boxes can be separated by variable spacer segments and many KRAB proteins contain only the A box.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
