Test Principale: This assay employs a two-site sandwich ELISA to quantitate PABPN1 in samples. An antibody specific for PABPN1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPABPN1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PABPN1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PABPN1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: PABPN1 encodes an abundant nuclear protein that binds with high affinity to nascent poly (A) tails. The protein is required for progressive and efficient polymerization of poly (A) tails on the 3' ends of eukaryotic genes and controls the size of the poly (A) tail to about 250 nt. At steady-state, this protein is localized in the nucleus whereas a different poly (A) binding protein is localized in the cytoplasm. An expansion of the trinucleotide (GCG) repeat from normal 6 to 8-13 at the 5' end of the coding region of this gene leads to autosomal dominant oculopharyngeal muscular dystrophy (OPMD) disease. Multiple splice variants have been described but their full-length nature is not known. One splice variant includes introns 1 and 6 but no protein is formed.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
