Test Principale: This assay employs a two-site sandwich ELISA to quantitate P4HA2 in samples. An antibody specific for P4HA2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyP4HA2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for P4HA2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of P4HA2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: Prolyl 4-hydroxylase catalyzes the posttranslational formation of 4-hydroxyproline in collagens. The vertebrate enzyme is a tetramer of 2 alpha subunits and 2 beta subunits. The beta subunit (P4HB) is an unusual multifunctional polypeptide identical to protein disulfide isomerase. P4HA2 is one of at least 2 alpha subunit isoforms. This gene encodes a component of prolyl 4-hydroxylase, a key enzyme in collagen synthesis composed of two identical alpha subunits and two beta subunits. The encoded protein is one of several different types of alpha subunits and provides the major part of the catalytic site of the active enzyme. In collagen and related proteins, prolyl 4-hydroxylase catalyzes the formation of 4-hydroxyproline that is essential to the proper three-dimensional folding of newly synthesized procollagen chains.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
