Test Principale: This assay employs a two-site sandwich ELISA to quantitate POP5 in samples. An antibody specific for POP5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPOP5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for POP5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of POP5 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: POP5. The deduced 163-amino acid protein has a calculated molecular mass of 18.8 kD. Western blot analysis of HeLa cells detected endogenous POP5 at an apparent molecular mass of 19 kD. HeLa cell POP5 associated with RNase MRP and RNase P RNAs. The immunoprecipitated RNase P complex was able to cleave pre-tRNA into mature tRNA and its 5-prime leader sequence, indicating that POP5 is part of the catalytically active RNase P complex. Mutation analysis showed that the C terminus of POP5 was not required for association with RNase MRP or RNase P complexes or for RNase P activity.
