Test Principale: This assay employs a two-site sandwich ELISA to quantitate SEPT6 in samples. An antibody specific for SEPT6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySEPT6 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SEPT6 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SEPT6 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: Septin-6 is a member of the septin family of GTPases. Members of this family are required for cytokinesis. One version of pediatric acute myeloid leukemia is the result of a reciprocal translocation between chromosomes 11 and X, with the breakpoint associated with the genes encoding the mixed-lineage leukemia and septin 2 proteins. This gene encodes four transcript variants encoding three distinct isoforms. An additional transcript variant has been identified, but its biological validity has not been determined. The deduced 424-amino acid protein shares 40% identity with CDC10 (SEPT7) and contains an ATP/GTP-binding site motif. Northern blot analysis detected nearly ubiquitous expression, with highest level in thymus.
