Application: This Human SH3 domain-binding glutamic acid-rich-like protein (SH3BGRL) ELISA Kit employs a two-site sandwich ELISA to quantitate SH3BGRL in samples. An antibody specific for SH3BGRL has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySH3BGRL present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SH3BGRL is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SH3BGRL bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Proline-rich peptide sequences have been shown to play important roles in protein-protein interactions that occur in signal transduction pathways. Specific proline-rich consensus sequences that bind SH3 domains have been identified. The predicted 114-amino acid SH3BGRL protein has a potential cell attachment sequence. A proline-rich sequence containing an SH3-binding motif is present in the middle of the protein. The SH3BGRL protein shares 60% identity with the proline-rich central region of SH3BGR. Northern blot analysis detected a single SH3BGRL transcript of approximately 1.9 kb that was present in all tissues examined.Mouse cDNA encoding a deduced protein that is 95% identical to human SH3BGRL.
