Test Principale: This assay employs a two-site sandwich ELISA to quantitate CD163 in samples. An antibody specific for CD163 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCD163 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CD163 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CD163 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview: CD163, a hemoglobin scavenger receptor, is expressed in monocytes and macrophages.Acute phase-regulated receptor involved in clearance and endocytosis of hemoglobin/haptoglobin c by macrophages and may thereby protect tissues from free hemoglobin-mediated oxidative damage.CD163 may play a role in the uptake and recycling of iron, via endocytosis of hemoglobin/haptoglobin and subsequent breakdown of heme. It binds hemoglobin/haptoglobin complexes in a calcium-dependent and pH-dependent manner. It exhibits a higher affinity for complexes of hemoglobin and multimeric haptoglobin of HP*1F phenotype than for complexes of hemoglobin and dimeric haptoglobin of HP*1S phenotype.
Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
