Test Principale: This assay employs a two-site sandwich ELISA to quantitate SPON2 in samples. An antibody specific for SPON2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySPON2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SPON2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SPON2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview: The full-length SPON2 cDNA encodes a 331-amino acid protein with a domain organization similar to those of zebrafish mindin-1/mindin-2 and F-spondin (SPON1) : a hydrophobic signal sequence in the N terminus, an FS1 domain, an FS2 domain, and a thrombospondin type I repeat. The mouse mindin protein, which is 85% identical to the human protein, is a conserved member of a family of secreted extracellular matrix proteins. RNA blot analysis detected expression in a variety of tissues, with abundant expression in lung and lymphoid tissues. Immunoblot blot analysis showed expression of a 42-kD protein under reducing conditions and expression of dimers and oligomers in a concentration-dependent manner under nonreducing conditions.
Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
