Application: This Human Transient receptor potential cation channel subfamily V member 3 (TRPV3) ELISA Kit employs a two-site sandwich ELISA to quantitate TRPV3 in samples. An antibody specific for TRPV3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTRPV3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TRPV3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TRPV3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

• Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
• It is recommended that all samples and standards be assayed in duplicate or triplicate.

Background: The TRPV3 protein belongs to a family of nonselective cation channels that function in a variety of processes, including temperature sensation and vasoregulation. The thermosensitive members of this family are expressed in subsets of sensory neurons that terminate in the skin, and are activated at distinct physiological temperatures. This channel is activated at temperatures between 22 and 40 degrees C. The gene lies in close proximity to another family member (TRPV1) gene on chromosome 17, and the two encoded proteins are thought to associate with each other to form heteromeric channels. The TRPV3 channel is widely expressed in the human body, especially in the skin in keratinocytes, but also in the brain. It functions as a molecular sensor for innocuous warm temperatures.
