Test Principale: This assay employs a two-site sandwich ELISA to quantitate TWSG1 in samples. An antibody specific for TWSG1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTWSG1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TWSG1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TWSG1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: The cDNA encodes a protein sharing 41% amino acid identity with Drosophila Tsg, 89% identity with the partial human Tsg sequence, and 94% identity with a mouse EST. The Tsg sequence contains a signal peptide, as expected for a secreted protein, and 2 conserved domains containing multiple cysteines at its amino and carboxy termini. At the late gastrula stage, maternal transcripts decrease and zygotic transcripts appear, specifically in the ventral region of the embryo. Tsg has ventralizing activity and is a bone morphogenetic protein (BMP) binding protein. The N-terminal domain of Tsg is sufficient to interact with BMP4 but not with chordin. Tsg competes for binding of BMP4 with the first cysteine-rich domain of chordin (CR1) but not with full-length chordin. Endogenous Tsg antagonizes CR1 activity.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
