Large tumor suppressor 1 (LATS1) is a serine/threonine kinase that belongs to the AGC kinase family of proteins. It is a core component of the Hippo signaling pathway in mammals. MST1/2 together with SAV1 phosphorylates and activates LATS1/2 and MOB1, which in turn phosphorylate YAP1 and TAZ. Phosphorylation of YAP1 by LATS1/2 prevents its translocation to the nucleus and thereby inhibits cell proliferation, migration, and cell death. LATS1 also regulates other biological processes such as cell cycle progression, apoptosis, renewal, and differentiation of stem cells. It acts as a tumor suppressor; studies demonstrated that loss of LATS1, deficiency of LATS1, or mutation in LATS1 reduced YAP1 phosphorylation and promotes tumor growth in multiple cancers. Hence LATS1 could be a promising target for anti-cancer therapies. ’s LATS1 (Human) ELISA kit quantitatively measures LATS1 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of LATS1 in the sample or standard.
