Test Principale: This assay employs a two-site sandwich ELISA to quantitate LN in samples. An antibody specific for LN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyLN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LN is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LN bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: Laminin, a protein network foundation for most cells and organs. The laminins are an important and biologically active part of the basal lamina, influencing cell differentiation, migration, adhesion as well as phenotype and survival. Laminins are large trimeric proteins that contain an α-chain, a β-chain and a γ-chain, found in five, three and three genetic variants, respectively. The laminin molecules are named according to their chain composition. Thus, laminin-511 contains α5, β1 and γ1 chains. Fourteen other chain combinations have been identified in vivo. The trimeric proteins form a cross, giving a structure that can bind to other cell membrane and extracellular matrix molecules. The three shorter arms are particularly good at binding to other laminin molecules, which allows them to form sheets. The long arm is capable of binding to cells, which helps anchor organized tissue cells to the membrane.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
