Test Principale: This assay employs a two-site sandwich ELISA to quantitate PYHIN1 in samples. An antibody specific for PYHIN1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPYHIN1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PYHIN1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PYHIN1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: PYHIN1 belongs to the HIN200 family of interferon-inducible proteins that share a 200-amino acid signature motif at their C-terminal ends. HIN200 proteins are primarily nuclear and are involved in transcriptional regulation of genes important for cell cycle control, differentiation, and apoptosis . The IFIX-alpha-2, -beta-2, and -gamma-2 isoforms lack 9 amino acids in their N-terminal regions relative to IFIX-alpha-1, -beta-1, and -gamma-1. IFIX-alpha and -beta isoforms have the type A C-terminal 200-amino acid signature motif of HIN200 proteins, including the conserved sequence MFHATVAT, whereas the IFIX-gamma isoforms lack the 200-amino acid signature sequence. The common N-terminal region of IFIX contains a predicted pyrin (MEFV) protein-protein interaction domain and a nuclear localization signal.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
