Test Principale: This assay employs a two-site sandwich ELISA to quantitate PDHX in samples. An antibody specific for PDHX has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPDHX present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PDHX is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PDHX bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: The PDHX gene encodes component X of the pyruvate dehydrogenase (PDH) complex. For a detailed description of the pyruvate dehydrogenase complex. The mammalian PDH complex differs from that in E. coli and from the other mammalian alpha-keto acid dehydrogenases by the presence of a 53-kD protein called protein X. Component X binds to the E3 component of the PDH complex. The open reading frame encodes an 501-amino acid protein sequence that is 20% identical with that encoded by the yeast PDX1 gene and 40% identical with that encoded by the lipoacyltransferase component E2 of the PDH (E2), and includes a lipoyl-bearing domain. Sequence comparisons showed that the PDHX protein contains an E3-binding domain.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
