Application: This Mouse Transmembrane 4 L6 family member 1 (TM4SF1) ELISA Kit employs a two-site sandwich ELISA to quantitate TM4SF1 in samples. An antibody specific for TM4SF1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTM4SF1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TM4SF1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TM4SF1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

• Unused wells must be kept desiccated at 4 °C in the sealed bag provided.
• Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
• It is recommended that all samples and standards be assayed in duplicate or triplicate.

Background: TM4SF1 is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is a cell surface antigen and is highly expressed in different carcinomas. The predicted peptide sequence was 202 amino acids long and contained 3 predicted NH2-terminal hydrophobic transmembrane regions which were followed by a hydrophilic region containing 2 potential N-linked glycosylation sites and a COOH-terminal hydrophobic transmembrane region.
