Application: This Mouse Tumor necrosis factor receptor superfamily member 11A (TNFRSF11A) ELISA Kit employs a two-site sandwich ELISA to quantitate TNFRSF11A in samples. An antibody specific for TNFRSF11A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTNFRSF11A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNFRSF11A is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNFRSF11A bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: RANk is a member of the TNF-receptor superfamily. This receptors can interact with various TRAF family proteins, through which this receptor induces the activation of NF-kappa B and MAPK8/JNK. This receptor and its ligand are important regulators of the interaction between T cells and dendritic cells. This receptor is also an essential mediator for osteoclast and lymph node development. Studies of the mouse counterpart suggest that this receptor directly mediates the osteoprotegerin ligand (OPGL) -induced osteoclastogenesis in osteoclast precursor cells. Northern blot analysis indicated that the 4.5-kb human RANK mRNA is expressed ubiquitously. In some human cell lines, the authors detected additional transcripts that were derived from the use of alternative polyadenylation signals in the RANK gene.
