Description: Peptidase M20 domain containing 1 (PM20D1), also known as N-fatty-acyl-amino acid synthase/hydrolase, is a secreted enzyme that catalyzes the biosynthesis of N-fatty acyl amino acids (NAAs) from free amino acids and free fatty acids and also catalyzes the reverse hydrolysis reaction. NAAs play an important role in various physiological functions as lipid signaling molecules. Additionally, they can stimulate oxidative metabolism via mitochondrial uncoupling. Mouse and human PM20D1 share about 71% amino acid identity. PM20D1 is involved in energy homeostasis. Ablation of PM20D1 in mice has shown bidirectional dysregulation of circulating NAAs, insulin resistance, altered body temperature, glucose intolerance etc. PM20D1 has also been associated with Alzheimer’s disease. Therefore, studying PM20D1 activity is of great interest in treating obesity, obesity-related disorders and neurological diseases. ’s PM20ctivity Assay Kit is a simple, rapid and sensitive assay to detect the activity of PM20D1 in various sample types. In this assay, PM20D1 catalyzes the conversion of N-Oleoyl-Glycine to generate glycine. In the subsequent steps, glycine in the presence of converter and developer generate hydrogen peroxide, which reacts with the PM20D1 probe to generate color measured at OD 570 nm. The assay can detect less than 0.1 mU under the assay conditions.
