Application: This Rat Anti-alpha-Fodrin IgG/IgA (A-Fodrin-IgG/IgA) ELISA Kit employs the competitive enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to A-Fodrin-IgG/IgA. Standards or samples are then added to the appropriate microtiter plate wells with a Horseradish Peroxidase (HRP) -conjugated A-Fodrin-IgG/IgA and incubated. The competitive inhibition reaction is launched between with HRP labeled A-Fodrin-IgG/IgA and unlabeled A-Fodrin-IgG/IgA with the antibody. A substrate solution is added to the wells and the color develops in opposite to the amount of A-Fodrin-IgG/IgA in the sample. The color development is stopped and the intensity of the color is measured.

• It is recommended that all samples and standards be assayed in duplicate or triplicate.

Background: Fodrin (also named nonerythroid spectrin) is a universally expressed membrane-associated cytoskeletal protein consisting of alpha- and beta-subunits. Fodrin is important for maintaining normal membrane structure and supporting cell surface protein function. Alpha-fodrin is one of the primary targets cleaved by caspases during apoptosis. The full length 240 kDa protein can be cleaved at several sites within its sequence by activated caspases to yield amino-terminal 150 kDa, carboxy-terminal 120 kDa and 35 kDa major products. Cleavage of alpha-fodrin leads to membrane malfunction and cell shrinkage.
