Application: This Rat C-C motif chemokine 6 (CCL6) ELISA Kit employs a two-site sandwich ELISA to quantitate CCL6 in samples. An antibody specific for CCL6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCCL6 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CCL6 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CCL6 bound in the initial step. The color development is stopped and the intensity of the color is measured.

• Allow all reagents to warm to room temperature for at least 30 minutes before opening.
• Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
• Unused wells must be kept desiccated at 4 °C in the sealed bag provided.
• Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
• It is recommended that all samples and standards be assayed in duplicate or triplicate.

Background: C10, known also as SCYA6 (small inducible cytokine A6) is one of several mRNAs that are acutely expressed in myelopoietic mouse bone marrow cultures stimulated by GM-CSF. It is also strongly elevated during the induction of neutrophilic differentiation of 32D cells that are dependent on the presence of IL3 or G-CSF.C10 is related to MIP-1. Its protein sequence demonstrates that C10 belongs to the family of chemotactic cytokines known as Chemokines (beta-Chemokines or CC-Chemokines) . The protein contains six instead of four conserved cysteine residues. The C10 gene contains a second exon not found in other Chemokines. This exon encodes a 16 amino acid sequence, which is inserted into the amino terminal region of the protein and plays a key role in recognizing or activating chemokine receptors.
