Application: This Rat Double-strand break repair protein MRE11A (MRE11A) ELISA Kit employs a two-site sandwich ELISA to quantitate MRE11A in samples. An antibody specific for MRE11A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMRE11A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MRE11A is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MRE11A bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: MRE11 encodes a nuclear protein involved in homologous recombination, telomere length maintenance, and DNA double-strand break repair. By itself, the protein has 3' to 5' exonuclease activity and endonuclease activity. The protein forms a complex with the RAD50 homolog; this complex is required for nonhomologous joining of DNA ends and possesses increased single-stranded DNA endonuclease and 3' to 5' exonuclease activities. In conjunction with a DNA ligase, this protein promotes the joining of noncomplementary ends in vitro using short homologies near the ends of the DNA fragments. MRE11 has a pseudogene on chromosome 3. Alternative splicing of MRE11 results in two transcript variants encoding different isoforms.
