Test Principale: This assay employs a two-site sandwich ELISA to quantitate CXCL5 in samples. An antibody specific for CXCL5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCXCL5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CXCL5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CXCL5 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: ENA-78 is a CXC chemokine that was originally isolated from media conditioned by the growth of a mouse lung type-II alveolar epithelial cell line (A549) stimulated by IL-1 or TNF. The full-length cDNA encodes a 114 amino acid (aa) residue precursor protein with a 36 aa residue signal peptide that is cleaved to generate the 78 aa residue secreted protein. ENA-78 shares significant amino acid sequence identity with NAP-2 (53%), GRO, , and (52%, 48% and 51%, respectively), and IL-8 (22%) . The gene for ENA-78 has been mapped to chromosome 4q13-q21. Like other CXC chemokines, ENA-78 is a neutrophil attractant and activator in vitro. Based on cross-desensitization experiments, it has been suggested that ENA-78 activity can be mediated through the IL-8 receptor system.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
