Background: RUNX2 (Runt-related transcription Factor) is a transcription factor that binds to its canonical consensus sequence 5’- PYGPYGGT-3’ in a number of promoters and enhancers. They include LCK, T-cell receptor enhancers, and various bone related genes such as osteocalcin, osteopontin, and bone sialoprotein. As a result of this, RUNX2 is involved in osteoblastic differentiation and skeletal morphogenesis. RUNX2 contains a proline/serine/ threonine-rich region at its C-terminus that has shown to be critical for its transcriptional activity.
