Soluble RAGE (sRAGE) is a free circulating isoform of RAGE that contains extracellular domains but lacks transmembrane and intracytoplasmic regions. sRAGE is formed either by proteolytic cleavage of RAGE by matrix metalloproteinases and ADAM-10 or by alternative splicing of endogenous secretory RAGE (esRAGE). It acts as a decoy of RAGE, prevents the binding of extracellular AGEs to cell surface RAGE, and thereby inhibits AGE/RAGE signaling. Therapeutic treatment of sRAGE demonstrated beneficial effects in diabetic atherosclerosis, polycystic kidney disease, autoimmune myocarditis, amyotrophic lateral sclerosis (ALS), and cancer. ’s sRAGE (Human) ELISA kit quantitatively measures sRAGE in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of sRAGE in the sample or standard.
