Stimulator of interferon genes (STING1, TMEM173) is a transmembrane protein located in the endoplasmic reticulum. The main function of STING1 is to protect the host from invading pathogens such as HIV, Plasmodium, and Mycobacterium tuberculosis. STING1 is expressed in T-lymphocytes, NK cells, monocytes, and myeloid cells. High expression of STING1 has been observed in several tissues such as the lung, heart, smooth muscle, retina, and ovary. Upon pathogen infection, STING1 detects double-stranded nucleic acids and induces downstream Type I interferon signaling. STING1 can also directly bind to bacterial cyclic dinucleotides such as cyclic di-GMP, promote the production of proinflammatory cytokines and subsequently eliminate bacteria. Mutations in STING1 lead to an autoinflammatory disease called STING-associated vasculopathy with onset in infancy (SAVI) that is characterized by systemic inflammation, interstitial lung disease, and recurring bacterial infection. ’s STING1 (Human) ELISA kit quantitatively measures STING1 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of STING1 in the sample or standard.
