Translocator protein (TSPO, PBR) is an 18 kDa highly conserved five transmembrane domain (TM) protein that belongs to the family of tryptophan-rich sensory proteins. This peripheral benzodiazepine receptor (PBR) protein is present on the outer mitochondrial membrane and is predominantly expressed in glial cells in the brain. Other tissues that express TSPO include the liver, heart, testis, and adrenal gland. TSPO performs multiple functions; it translocates cholesterol from the outer to the inner mitochondrial membrane that is a rate-limiting step in steroid and bile acid synthesis. TSPO plays a protective role in the heart; it regulates the rate and contractility of the heart and controls inflammatory response during infections. TSPO is upregulated in glial cells during neural disorders and injury and thus serves as an efficient neuroinflammation marker. Several ligands of TSPO are currently under investigation for the treatment of neurological and psychiatric disorders. ’s TSPO (Human) ELISA kit quantitatively measures TSPO in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of TSPO in the sample or standard.
