Description: Acetyl-CoA is a component of cellular respiration that adds acetyl groups to biochemical reactions. These reactions are used in the metabolizing of proteins, carbohydrates, and lipids that will provide energy sources in the forms of adenosine triphosphate, lactic acid, and ketone bodies. Acetyl-CoA is synthesized in mitochondria by a number of reactions: oxidative decarboxylation of pyruvate; catabolism of some amino acids and β-oxidation of fatty acids. Its main function is to deliver the acetyl group to the citric acid cycle (Krebs cycle) to be oxidized for energy production. Acetyl-CoA is a component of cellular respiration (energy conversion) that adds acetyl groups to biochemical reactions. Acetyl-CoA levels reflect energy production potential in the disease state. ’s Acetyl-CoA ELISA Kit is used to quantitatively measure Acetyl-CoA in Serum, plasma and other biological fluids. The kit is based on the Sandwich ELISA priple. The micro-ELISA plate provided in this kit has been pre-coated with an antibody specific to Acetyl-CoA. Samples (or Standards) are added to the micro-ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Acetyl-CoA and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Acetyl-CoA, biotinylated detection antibody and Avidin HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm. The OD value is proportional to the concentration of Acetyl-CoA. You can calculate the concentration of Acetyl-CoA in the samples by comparing the OD of the samples to the standard curve. The concentration of Acetyl-CoA in the samples is calculated by comparing the OD of the samples to the standard curve.
