Description: ADAM28 is a member of the ADAM (a disintegrin and metalloproteinase) gene family and have been implicated in a variety of biological processes involving cell–cell and cell–matrix interactions, including fertilization, muscle development, and neurogenesis. It consists of two isoforms, prototype membrane-type form and short secreted form. The metalloproteinase domain of ADAM28 has the zinc-binding consensus sequence, and ADAM28 exhibits catalytic activity to a few substrates such as insulin-like growth factor binding protein-3. It plays a role in the adhesive and proteolytic events that occur during lymphocyte emigration or may function in ectodomain shedding of lymphocyte surface target proteins, such as FASL and CD40L. In human non-small cell lung carcinomas and breast carcinomas, ADAM28 is overexpressed predominantly by carcinoma cells, and the expression correlates with carcinoma cell proliferation and lymph node metastasis. A8 acts as a target for cancers. ’s ADAM28 (Human) ELISA Kit is used for the quantitative detection of ADAM28 in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of ADAM28 in the sample.
