Alpha-2-macroglobulin is a large plasma protein mainly produced by the liver. It inhibits a variety of proteinases and binds several types of ligands such as growth factors and cytokines. It stimulates the regeneration of lymphocytes in irradiated mice, aids in the transport of zinc and controls the clotting and fibrinolytic system. It has been localized to diffuse amyloid plaques in Alzheimer’s disease patients. The levels of Alpha-2-macroglobulin are increased in liver cirrhosis, nephrotic syndrome, diabetes, and severe burn cases. ’s Alpha-2-macroglobulin (Human) ELISA Kit is used for the quantitative detection of Alpha-2-macroglobulin in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of Alpha-2-macroglobulin in the sample.
