Unit Definition: One unit of BirA is defined as the amount of enzyme that catalyzes the biotinylation of 1 µmol of Avi-tagged protein (CD40L-Avi) per minute under the assay conditions, where 50% of CD40L-Avi is biotinylated in 60 min at 30 °C in the reaction buffer containing 5 mM MgCl2, 2 mM ATP, 150 µM biotin and 50 µM substrate in PBS and monitored on an SDSPAGE.
