Test Principale: This assay employs a two-site sandwich ELISA to quantitate TOMM40L in samples. An antibody specific for TOMM40L has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTOMM40L present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TOMM40L is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TOMM40L bound in the initial step. The color development is stopped and the intensity of the color is measured.

TOMM40 is the channel-forming subunit of the translocase of the mitochondrial outer membrane (TOM) complex that is essential for protein import into mitochondria.Extraction and antibody-accessibility studies suggested that Tomm40 is embedded within the mitochondrial membrane.Tomm40 was associated with a 400-kD complex containing the mitochondrial import receptors Tom20 and Tom22 and several smaller components. Mitochondria-targeted preproteins interacted with the Tomm40-containing complex during their transit to the mitochondrial matrix, and immunodepletion of Tomm40 caused the loss of preprotein import activity.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
