Test Principale: This assay employs a two-site sandwich ELISA to quantitate TNP1 in samples. An antibody specific for TNP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTNP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNP1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNP1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: Transition protein-1 is a spermatid-specific product of the haploid genome which replaces histone and is itself replaced in the mature sperm by the protamines. Yu et al. (2000) generated mice lacking the major transition nuclear protein, TP1, by targeted deletion of the Tnp1 gene in mouse embryonic stem cells. Surprisingly, testis weights and sperm production were normal in the mutant mice, and only subtle abnormalities were detected in sperm morphology. Electron microscopy showed large rod-like structures in the chromatin of mutant step 13 spermatids, in contrast to the fine chromatin fibrils observed in wildtype. Steps 12-13 spermatid nuclei from the testis of Tnp1-null mice contained, in place of TP1, elevated levels of TP2 (TNP2) and some PRM2 precursor.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
