Host: Mouse
Reactivity: Human
Homology: N/A
Immunogen: Recombinant human protein was used as the immunogen for the CD146 antibody.
Research Area: Cancer, Immunology, Signal Transduction
Tested Application: Flow, IF, IHC-P
Application: Flow Cytometry: 0.5-1 ug/million cells in 0.1ml
Immunofluorescence: 1-2 ug/ml
Immunohistochemistry (FFPE) : 1-2 ug/ml for 30 min at RT (1)
Prediluted format : incubate for 30 min at RT (2)
Titering of the CD146 antibody may be required for optimal performance.
1. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 10-20 min followed by cooling at RT for 20 minutes
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required) , drip mAb solution onto the tissue section and incubate at RT for 30 min.

Alternate Name: Cell surface glycoprotein MUC18, Cell surface glycoprotein P1H12, Melanoma cell adhesion molecule, Melanoma-associated antigen A32, Melanoma-associated antigen MUC18, S-endo 1 endothelial-associated antigen, CD146, MCAM, MUC18
User Note: Optimal dilutions for each application to be determined by the researcher
BACKGROUND: CD146/MUC18/MCAM plays a role in cell adhesion, and in cohesion of the endothelial monolayer at intercellular junctions in vascular tissue. Its expression may allow melanoma cells to interact with cellular elements of the vascular system, thereby enhancing hematogeneous tumor spread. Could be an adhesion molecule active in neural crest cells during embryonic development. Acts as surface receptor that triggers tyrosine phosphorylation of FYN and PTK2/FAK1, and a transient increase in the intracellular calcium concentration. [UniProt]
