Test Principale: This assay employs a two-site sandwich ELISA to quantitate UGT8 in samples. An antibody specific for UGT8 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyUGT8 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for UGT8 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of UGT8 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: Galactocerebrosides are abundant sphingolipids of the myelin membrane of the central nervous system and peripheral nervous system and are also present in small amounts in kidney. The key enzymatic step in the biosynthesis of galactocerebrosides consists of the transfer of galactose to ceramide catalyzed by UDP-galactose ceramide galactosyltransferase (CGT) . The enzyme encoded by the CGT gene is the first involved in complex lipid biosynthesis in the myelinating oligodendrocyte.The same part of the predicted amino acid sequence that binds the ceramide moiety binds aglycone in isozymes of the UGT subfamilies. Only subtle changes of the rather conserved C-terminal domain appeared to be sufficient to confer the specificity of the binding site for the activated sugar donor molecule UDP-galactose.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
