Test Principale: This assay employs a two-site sandwich ELISA to quantitate ACSF2 in samples. An antibody specific for ACSF2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyACSF2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ACSF2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ACSF2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview: Fatty acids are incorporated into membranes and signaling molecules and have roles in energy storage and metabolism. These essential functions require activation of the fatty acid by acyl-coenzyme A (CoA) synthetases, such as ACSF2, which form an activating thioester linkage between the fatty acid and CoA.The deduced 615-amino acid protein contains all 5 motifs characteristic of acyl-CoA synthetases. Phylogenetic analysis revealed that ACSF2 belongs to a small group of acyl-CoA synthetases that differ significantly from all other clades of acyl-CoA synthetases and from each other. FLJ20920 contains a PPARG-recognition element in its promoter sequence. Transfection of antisense FLJ20920 into preadipocytes resulted in decreased triglyceride accumulation and reduced expression of GLUT4 (SLC2A4) and HSL (LIPE) mRNA.

Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
